Note (General): 少量のMethamphetamine(METH)を反復投与し、中脳に小胞体ストレス蛋白の増加を認めるモデル動物に対して多量のMETHを投与した。この動物の中脳の解析により、小胞体ストレス蛋白のGrp78および活性化ATF6の有意な増加を認めた。さらに、酸化ストレスのマーカーである4-HNEの増加の抑制や抗酸化ストレス蛋白であるSOD1の早期の増加を認めた。また、アポトーシスの指標が抑制され、少量のMETH反復前投与は、多量のMETH投与における中脳の神経細胞に対する感受性に影響することが示唆された。
We previously reported that low-dose methamphetamine (METH) pre-treatment increases endoplasmic reticulum stress protein levels in the rat midbrain. In this study, we assessed the effects of high-dose METH on rat midbrain. High-dose METH administration led to a significant increase in expression of the endoplasmic reticulum stress protein Grp78 and activated ATF6. Furthermore, we observed an early increase of the antioxidant stress protein SOD1 and increased inhibition of the oxidative stress marker 4-HNE. High-dose METH inhibited the apoptotic index in midbrain. Collectively, these results suggest that low-dose METH pre-treatment influenced the susceptibility of midbrain neurons to high-dose METH administration.
