蛍光プローブ標識による分子拡散計測の信頼性を原子分解能を有する原子間力顕微鏡(AFM)を用いた直接計測で評価するために、AFM装置および周辺機器の開発を行った。AFM試料ホルダに光学窓や溶液フロー機能を組み込み、周波数変調AFM(FM-AFM)により脂質二重層上に存在するサブナノメートルの表面形状を直接観察できることを示した。この試料ホルダは落射型蛍光顕微鏡による観察を考慮した設計となっており、FM-AFM計測との比較により蛍光プローブ分子拡散計測法の信頼性を評価できるようになった。
In this project, an atomic force microscopy (AFM) instrument was developed to evaluate the reliability of fluorescence probe methods for molecular diffusion measurements in lipid bilayers. A sample holder developed in this project is usable in both the molecular-resolution AFM imaging and fluorescence measurements under the same condition. As the developed AFM instrument allows us to compare the data of molecular diffusion obtained by both methods, the capability should be useful for the reliability evaluation of fluorescence probe methods for molecular diffusion measurements.
