肝臓でのストレス応答が、肝臓インスリン抵抗性および糖代謝異常に果たす役割については、必ずしも十分に解明されていない。肥満・インスリン抵抗性状態や小胞体ストレス誘導時において、ストレス誘導性転写因子であるATF3発現の増加を確認した。またこの時、IL-6による糖新生系酵素遺伝子発現を抑制する転写因子STAT3のリン酸化は減弱していた。これらの結果は、肥満・インスリン抵抗性時の肝臓糖代謝異常にATF3やSTAT3が何らかの役割を果たす可能性を示している。
The role of hepatic stress response in insulin resistance and glucose metabolism is not elucidated enough. Through this study, we have clarified expression of ATF3, is transcription factor induced by stress, increased in the condition of obesity and insulin resistance and the induction of endoplasmic reticulum. Then, phosphorylation of STAT3, inhibits the expression of glucose production related gene in the liver, by IL-6 was decreased. These results suggest that there is possibility that ATF3 and STAT3 in the liver regulate the glucose metabolism.
