大腸癌培養細胞を用いた研究で，ZFP57を過剰発現させると，その増殖能が亢進するとともに，マウスの脾注モデルで大腸癌肝転移を促進することを明らかにした．一方，ZFP57をノックダウンすると増殖能が抑制されるとともに，マウスにおける肝転移が抑制された．大腸癌の臨床検体を用いてZFP57の発現を免疫組織学的に評価した結果，肝転移を有する症例の原発巣や肝転移巣では肝転移陰性例の原発巣に比べてZFP57が過剰発現している頻度が有意に高く，多変量解析でもZFP57の過剰発現が肝転移の有意な規定因子と判断された．以上より，ZFP57過剰発現は大腸癌肝転移を促進することが明らかとなった．
In liver metastatic experiments using human colorectal cancer cell lines, liver polymetastasis occurred with high frequency in the case of ZFP57-overexpressing cells, while control cells produced only oligometastasis. When we examined the relationship between the clinicopathological feature of colorectal cancer and ZFP57 overexpression using clinical specimens, liver metastasis-positive cases were more frequently associated with ZFP57 overexpression than negative cases in primary lesions of colorectal cancer. In addition, the expression level of ZFP57 showed significant correlations with that of NANOG, a metastasis-related gene. We also found that ZFP57 overexpression results in reduced disease-free survival rate of patients in colorectal cancer.This study demonstrated that ZFP57 plays an important role in liver metastasis of colorectal cancer. ZFP57 would be a novel target molecule of treatment for liver metastasis of colorectal cancer.
