C57BL/6Jマウスに高脂肪食を摂取させることで耐糖能異常を有するマウスモデルを作製した。次に、C57 BL/6Jマウスの皮下に、マウス膵臓癌細胞株PAN02を接種することで担癌状態マウスモデルの作製に成功した。この担癌状態マウスモデルの宿主免疫状態の特徴につき、マウス膵臓癌細胞株皮下接種後、4週間後および8週間後と経時的に末梢血液および脾細胞の炎症細胞の特徴および経時的な変化をFlow cytometryにて検討した。末梢血液および脾細胞にて抑制系の単球/マクロファージ系細胞であるMDSCsと言われる細胞集団の著明な増加を認め、腫瘍系の増大に一致して経時的に増加していた。
We could establish a murine model of impaired glucose tolerance.Next, We also could establish a murine model of pancreatic ductal adenocarcinoma model. We disclosed that frequency of myeloid derived suppressor cells was increased along increment of tumor size in this tumor bearing mice.
