mTOR阻害剤であるTemsirolimusは悪性胸膜中皮腫細胞株の増殖抑制効果を濃度依存性に示すことをIn Vitroで示した。特にp70s6kのリン酸化を阻害すること、またアポトーシスのマーカーとして知られるcleaved-caspase3の発現レベルを亢進させることを確認した。FACS 法により、MSTO211H以外の細胞株、H226,H290,and Y-MESO14ではアポトーシスの誘導が確認された。さらにTemsirolimusとCisplatinは相乗効果を示すこと、Temsirolimusは抗血管新生効果、特にVEGF発現抑制効果を示すことを明らかにした。Temsirolimusは抗腫瘍効果と、血管新生抑制効果の両面の作用を持つことが示された。
The results suggest the therapeutic efficacy of temsirolimus against MPM in vitro. And it is expected that temsirolimus inhibits MPM growth not only by anticancer effect but by antangiogenic effect in vivo.
