様々な細胞の代謝センサーであるATP感受性カリウムチャネル（KATPチャネル）を欠損させたマウスにおける、脂肪組織インスリン依存的糖取り込みの亢進について解析を進めた。欠損マウスの脂肪組織へのインスリン刺激により、Aktと未知タンパク質との結合が誘導されることを見出した。活性化Aktと未知タンパク質との結合は、インスリン濃度依存的に増大した。欠損マウス脂肪細胞内インスリン応答には、Akt2が主に寄与することが示された。マス解析により未知タンパク質の同定を試み、14-3-3タンパク質ファミリーがその候補として挙げられた。
ATP-sensitive potassium (K(ATP)) channel is critical in metabolism control. Previous reports showed that insulin-dependent glucose uptake is enhanced in adipose tissues of KATP channel knockout (KO) mice compared with that in wild-type mice. Insulin stimulated in vivo interaction of Akt and unknown factor in adipose tissues. The interaction was enhanced dependent on amount of injected insulin. Akt2 mainly played role for the interaction in adipose tissues of KO mice. Mass spectrometry analysis showed 14-3-3 family proteins are candidates for the interaction with Akt2.
