Note (General): 本研究は、悪性黒色腫発生および腫瘍が浸潤・転移していく過程におけるBRAF変異遺伝子とTWISTの発現の関連を調べた。TWISTは脱メチル化された状態で、mRNAレベル、蛋白レベルでともに高度に発現していた。BRAF変異遺伝子の有無とTWISTmRNAレベル、蛋白レベルの発現量との関連は認められなかった。原発腫瘍と転移性腫瘍においてTWISTmRNAレベルに明らかな違いは認めなかったが、蛋白レベルでは転移腫瘍でTWIST発現量が高い傾向が見られた。原発腫瘍において転移をきたした例と転移を認めていない例ではTWIST発現量に明らかな違いは認めなかった。以上の結果から悪性黒色腫ではTWISTのpromotor領域は脱メチル化されTWISTが比較的高頻度に発現されている。TWISTは転移を促進する方向へ作用するが、悪性黒色腫は転移をきたしやすい腫瘍であり、その転移腫瘍でTWISTの高発現を認めた今回の結果は悪性黒色腫の高転移能を説明すると思われる、ただし良性腫瘍の色素性母斑でもTWISTの発現が見られており、TWISTの発現以外に何らかの調節機構が働いていると思われる。
