Note (General): 本研究では患者の血液検体に、GFP遺伝子を組み込んだテロメレース特異的増殖型アデノウイルス(OBP-401)を感染させ、GFP発光する細胞を検出することで簡便に血中の循環癌細胞(Circulating Tumor Cell: CTC) を可視化して検出する技術を開発し、これを卵巣癌の再発モニタリング等に応用することを目的とした。偽陽性の主因である白血球による非特異的GFP発光を、CD45抗体２重染色にて除外する方法を確立し得た。実際の患者検体からも婦人科癌の約４０％にCTC検出を認めた。CTC検出は治療効果判定マーカーとなるのみならず、予後マーカーとしても有効であることが明らかとなった。
We established the novel detection system for circulating tumor cells (CTC) with telomerase-specific replication adenovirus in which GFP cDNA was integrated for GFP fluorescence. Infection of these viruses with 5ml of blood samples from the patients with gynecological cancers successfully identified CTC, while no CTC was detectd in the samples from healthy volunteers. While the number or the presence of CTCs were not significantly correlated with clinicopathological characteristics, they were tightly related to the treatment outcome as well as the overall survival. Thus, the present study clearly found the clinical utility of this novel detection system for CTC.
