Note (General): ヒトパピローマウイルス(HPV)感染病態における、発癌因子APOBECの発現誘導機構を解明した。HPV16(+)子宮頸部異形成由来細胞の分化により、APOBEC3A及びAPOBEC3Bの発現が誘導され、宿主ゲノムに変異が導入される事が示唆された(Wakae, Sci. Rep. 2018)。またHPV関連癌である子宮頸癌や中咽頭癌検体でも宿主ゲノム変異が導入され、APOBEC3Bの発現と相関した。HPV感染病態において、宿主細胞の分化がAPOBEC発現、及び宿主ゲノム変異を誘導する事を示した。
We clarified the mechanism how oncogenic host factors, APOBECs, are induced in HPV infected cells. We demonstrated that, differentiation of an HPV16(+) cervical keratinocyte induces APOBEC3A and 3B, as well as mutates host genome (Wakae, Sci. Rep. 2018). In addition, host gene mutations were detected from HPV-associated cancers, including cervical and oropharyngeal cancers, and were correlated with APOBEC3B expression. These results suggested that differentiation of HPV-infected host cells induces APOBECs and mutates their own genome.
