Note (General): 我々はCRISPR/Cas9システムを用いてSatb1の欠損細胞株を樹立した。Satb1欠損F12細胞にて，15q11-q13領域のクロマチン脱凝集状態を解析した所，正常細胞株と比較し脱凝集が小さくなっていることが明らかとなった。一方，Satb1を強制発現させたF12細胞では，クロマチン脱凝集状態が正常に比べ大きく広がっていることを見出した。このことから，15q11-q13領域のクロマチン脱凝集の構築には，Satb1が大きく関わっていることが明らかとなった。また， Satb1の欠損細胞株において，15q11-q13領域のMAGEL2/NDNの発現が低下していることが明らかとなった。
We established Satb1 deficient cell line using the CRISPR / Cas9 system. Analysis of chromatin disaggregation in the 15q11-q13 region of Satb1-deficient F12 cells revealed smaller disaggregation compared to normal cell lines. On the other hand, in F12 cells in which Satb1 was forcedly expressed, it was found that the chromatin disaggregation state spreads more than normal. From this, it became clear that Satb1 is greatly involved in the construction of chromatin disaggregation in the 15q11-q13 region. In addition, it was revealed that the expression of MAGEL2 / NDN in the 15q11-q13 region is reduced in the Satb1 deficient cell line.
