Note (General): 本研究では、灰化ウシ海綿骨を使った再生骨用担体に対し生体骨並みの力学的特性を持たせることを目標とし、以下の成果を得た。灰化ウシ海綿骨をナノ粒子化し、それとアルギン酸を混合した複合材を作製した。同複合材は、生体緻密骨並みの力学的剛性を有しており、さらにアルギン酸をシアノ―ル架橋することで疑似体液中での力学的安定性を確保することができた。加えて、ランダムパルス電磁場刺激とRGDSペプチド表面修飾を併用することにより担体内での骨芽細胞による基質産生と石灰化が促進されることがマイクロCT計測により確認した。
In this study, we aimed to give the mechano-compatibility to the scaffold for regenerated bone made of calcined bovine cancellous bone. The following results were obtained. Calcined bovine cancellous bone was nanoparticulated and mixed with alginic acid. The composite has mechanical properties close to those of a living compact bone. Furthermore, silanol crosslinking of the alginic acid made the composite mechanically-stable in simulated body fluid. In addition, Micro-CT imaging technique confirmed that the combination of random pulse electromagnetic field stimulation and surface modification with RGDS peptide promotes bone matrix production and calcification by osteoblasts in the scaffold.
