Note (General): 本研究では　Na+/H+交換輸送体(NHE)のうちNHE5の大腸癌における意義を解析しNHE5特異的阻害剤による新規抗がん治療法を開発する事を目的とした。癌組織では細胞外が酸性で細胞内がアルカリであり細胞内H+を細胞外に排出にNHEが深く関与している。NHE5は大腸癌組織において高頻度で過剰発現が確認され、NHE5が癌細胞における細胞外およびエンドゾーム内の弱酸性化環境形成に伴う癌細胞の細胞増殖能・浸潤能・転移能の獲得に重要な役割を果たしている可能性がある。NHE1阻害剤をリード化合物としてNHE5選択的阻害剤を分子設計合成し、NHE5活性の阻害効果と細胞増殖抑制効果を検討した。
Malignant tumors have an acidic extracellular pH and alkaline intracellular pH, which is facilitated by altered metabolism of tumor cells and the hypoxic tumor microenvironment. Na+/H+ exchangers (NHEs) play important roles in secreting protons in tumor cells. Neuron enriched NHE5 dynamically shuttles between the plasma membrane and recycling endosomes, serving as a mechanism that regulates pH across the endosomal and plasma membranes. NHE5 plays an important role in endocytic recycling of tyrosine kinase receptor. In this study, we try to establish a new therapeutic strategy for colon cancer.by synthesizing novel NHE5 specific inhibitor.
