Note (General): 癌局所での、血管外血小板凝集と腫瘍の増殖進展に関する分子細胞学的な事象に関してはほとんど明らかとなっていない。本研究では、ヒト血小板の膵癌細胞に及ぼす影響を検討した。健常人より得られた血小板と膵癌細胞株の共培養にて膵癌細胞の浸潤能の増加を認め、また癌抑制遺伝子であるPTENの発現低下を認めた。さらに膵癌細胞への接着に関してはCD44 standard formを有し、上皮間葉転換を認める細胞株に有意に接着した。またCD44 standard formを発現誘導することにより血小板の接着増加を認め、膵癌細胞と血小板接着、凝集にはCD44 standard formが関与することが示唆された。
Little is known about the molecular cytological events involved in extravasated platelet aggregation (EPA) and tumor progression in cancer microenvironment. In this study, we examined the effect of human platelets on pancreatic cancer cells. Co-culture of platelets obtained from healthy volunteers and pancreatic cancer cell lines showed an increase in invasion ability for pancreatic cancer cells, and also decreased in expression of PTEN, a tumor suppressor gene. Moreover, expression of CD44 standard form concerned to adhesion to pancreatic cancer cells for platelets. In addition, expression of the CD44 standard form increased the adhesion between platelets and pancreatic cancer cells, suggesting that CD44 standard form involved in pancreatic cancer cell and platelet adhesion.
