近年の蛍光タンパク質、および発光タンパク質を応用したイメージング技術の発展により、生体内での様々なバイオイメージングが可能となった。本研究では、透明な生物のモデル生物として、レプトケファルスを研究対象とし、透明性の分子基盤の解明を目指した。透明性とクロマチン高次構造との相関を探るために、細胞レベルで核の構造を解析した。ここでは、特にヘテロクロマチンの構造に注目するために、細胞をDAPI染色、およびヒストン修飾（H3K9me3など）で染色し、その核内での分布を、蛍光顕微鏡を用いて観察した。その結果、クロマチン高次構造と透明性との相関を見出すことに成功した。
Recent advantages of fluorescent proteins and chemi-luminescent proteins allow us to monitor various biological phenomenon through biological imaging. In this project, we aim to understand molecular mechanisms underlying transparency of Leptocephalus, transparent larva of the eel. We analyzed chromatin structures in cells of its transparent tissues to find out a contribution of chromatin structures onto the transparency. We first focused on heterochromatin structure, which is one of potential nuclear structure to affect reflection of light. We stained cells with DAPI and H3K9me3, a histone marker for heterochromatin and found no significant correlation between the transparency and three dimensional distribution of heterochromatin within nuclear space.
