ミネラル(必須微量元素)は活性酸素の生産や消去と密接に関連する因子であり、それらの輸送メカニズムを分子レベルで解明することは、細胞内における活性酸素濃度の制御メカニズムを理解するために必須である。本研究では、X線吸収スペクトルを利用し、活性酸素の生産に関連する鉄トランスポーターの局所構造を解析する手法を確立した。さらに、活性酸素の消去に関連するセレンのトランスポーターであるセレノプロテインPをヒト血漿から高純度に精製し、その微結晶を得ることに成功した。今後の結晶構造解析によりセレンの輸送メカニズム解明が期待される。
A fine-tuned balance between production and scavenging of reactive oxygen species (ROS) is crucial for living cells. Since minerals (essential trace elements) can be a factorin regulation of ROS level, mechanisms of mineral transport at molecular level are essential for understanding the tuning mechanism of the ROS level in living cells. In this study, techniques for analysis of local structure around mineral using X-ray absorption spectroscopy has been established. Also, micro crystals of selenoprotein P, a selenium transporter, have been obtained. Further crystallographic study will be contributed the finding of scavenging mechanism of ROS by selenium-sites in the protein.
