細胞ががん化する際には様々な代謝異常が生じるが、近年これらの代謝異常が、がん細胞の増殖や悪性化に深く関与していることが報告され、がん治療の新たな標的として注目を集めている。我々は、がん抑制遺伝子RBの機能喪失が、脂質代謝経路、特にメバロン酸経路の多くの遺伝子発現を誘導することを見出した。また、その結果生じるコレステロールやその誘導体脂質が、細胞内ROSレベルを抑制することにより、がんの悪性化形質の獲得に寄与していることを明らかにした。本研究は、RBによる脂質代謝制御という新たな機能と、その破綻が引き起こすメバロン酸経路の異常な亢進が、がんの悪性化形質の獲得に寄与していることを示すものである。
Recent advances in cancer metabolism have revealed that metabolic abnormalities in cancer cells are closely related to enhanced cell proliferation and malignant progression of a number of tumors.Mutations in Retinoblastoma (RB) gene are found at the initiation of limited human cancers, whereas RB protein inactivation is frequently found in much wider variety of cancers mainly during their progression. These findings suggest that RB exerts more roles than previously thought. Here we show that RB affects transcription of many enzymes involved in Mevalonate (MVA) pathway. We demonstrate that RB depletion in prostate cancer cells induces some of typical malignant features including tumor sphere forming-activity and hormone-refractory growth. Cholesterol and its derivatives generated by MVA pathway contribute to these phenotypes through modifying cellular ROS level.These results indicate that RB regulates cancer progression through the metabolic function in controlling MVA pathway.
