Note (General): 前立腺癌の特徴として造骨性骨転移の形成がある. 我々は癌骨細胞間の微少環境について研究計画を立案した.前立腺癌細胞LNCaP-SF をSCID マウスに接種することで前立腺再燃癌および前立腺癌骨転移の動物モデルを樹立し, トラニラスト(TRN)の抗腫瘍効果および作用機序について検討した.in vitro においてTRN は前立腺癌細胞の増殖を抑制し, 遺伝子, 蛋白レベルでアポトーシス関連因子の発現を誘導した. 骨関連細胞に対してはTGF-β1の産生を抑制した.In vivo において皮下への移植,脛骨への移植いずれもTRN 投与群は有意に腫瘍の増殖が抑制された. 以上の結果より, TRNには前立腺癌細胞に対するアポトーシスの誘導と,骨転移巣における間質細胞などに対するTGF-β1産生抑制とを介した抗腫瘍効果があることが示された.
