虚血再潅流傷害において、IL-6はTNF-αの上流に位置し、抗IL-6抗体を投与することによりTNF-α産生が抑制され、TNFα自体による皮膚傷害を軽減し、またTNFα産生に引き続いて生じるiNOSからのNO産生を抑制することにより皮膚障害を軽減する。従って、抗IL-6抗体は抗TNFα抗体よりも虚血再潅流傷害をより軽減できることが示唆された。またその投与方法としては、皮内注射が安全に投与できることが示唆されたが、理想的な投与方法である貼付ないし塗布は本モデルでは評価困難であり、虚血再潅流モデルの改良が必要であると考えられた。
In a mouse model of cutaneous ischemia-reperfusion injury, local administration of anti-IL-6 antibody inhibited expression of TNF-alpha and inducible nitric oxide synthase, and reduced cytotoxicity of them. And we checked the safeness about intradermal injection with anti-IL-6 antibody.From that reason, I suggest that anti-IL-6 antibody treatment might prevent the pressure ulcers.
