Note (General): 金沢大学がん進展制御研究所
当初SUCLA2のアイソザイムであるSUCLG2の阻害が合成致死性を発揮すると考えたが、その効果は、限定的であった。そこで、我々は、SUCLA2を欠く前立腺がん細胞株とそこにSUCLA2を再構成した前立腺がん細胞株を作成、CRISPR-CAS9ライブラリーを用い、SUCLA2を欠く前立腺がん細胞株において特異的にドロップアウトする代謝遺伝子を検索し、8の候補遺伝子を得た。さらに我々は、SUCLA2欠失前立腺がん特異的に細胞死を誘導する化合物のスクリーニングを行い、現在までに、天然化合物ライブラリーのスクリーニングから、化合物2種がSUCLA2欠失株特異的に細胞傷害性を示すことを明らかにした。
