カリウムチャネルサブユニット遺伝子KCNS3は、大脳皮質のパルブアルブミン陽性ニューロン（PVニューロン）に選択的に発現し、それによる周期的神経活動（オシレーション）の形成を促進することで脳内ネットワークにおける情報処理（認知機能）に役立っていると考えられる。統合失調症の患者で低下が認められる視覚情報の作業記憶は、前頭前野、外側頭頂葉、視覚野などからなる神経ネットワークにより支えられており、これらの領域ではオシレーションの異常が多く報告されている。本年度は、それぞれ領域のPVニューロンにおいてKCNS3の発現が低下していることを確かめるための死後脳サンプルの準備を行った。まず、前頭前野として46野、外側頭頂葉として7野、視覚野として17野および18野の組織をすべて得られる統合失調症例20と対照例20の死後脳組織を準備した。疾患群と対照群の間では、性別比は同じで（男:女=16:4）で、死亡時の平均年齢（疾患群45.6±9.5, 対照群47.7±9.6）、死後経過時間（疾患群15.5±5.8, 対照群14.4±6.2）がほぼ等しくなるように調整した。さらに、脳組織の品質を示すRNA　integrity number（疾患群8.3±0.5, 対照群8.2±6.5）やpH（疾患群 6.7±0.2, 対照群6.5±0.3）にも差はなく、良好な状態にあることが判明した。また、real-time PCRによる定量の条件決定を行い、視覚野から得られたRNAサンプルで、KCNS3遺伝子断片を99%の増幅効率にて内部標準遺伝子であるbeta-actin, cyclophylin, GAPDHと共に増幅・定量出来る条件を設定した。それぞれの症例の各領域からのRNAの抽出も行った。
