Note (General): 金沢大学学際科学実験センター
ヒメミカヅキモ、シャジクモは、陸上植物に近縁な藻類であり、ヒメミカヅキモは生活環を通じて単細胞性であるが、シャジクモはn世代だけが多細胞体制を作る。n, 2n両世代で多細胞体制を作る陸上植物への進化を解明するため、この2種でEST解析を進めた。昨年度決定したヒメミカヅキモNIES-67&68株栄養増殖ステージに加え、A2 ヒメミカヅキモNIES-67株(接合型プラス)性分化誘導ステージ;A3 ヒメミカヅキモNIES-68株(接合型マイナス)性分化誘導ステージ;の完全長cDNAライブラリーを作製し各1万9200クローンについて両端からのESTシークエンスを行った。これにより合計116, 694配列のESTが得られた。この配列を以前から得られていたESTとともにアセンブルし10 620個のcontigを得た。シャジクモについては、B2 シャジクモ生殖器官形成直前ステージ;B3 シャジクモ受精直前ステージの完全長cDNAライブラリーを作製し合計73, 388配列のESTが得られた。また、ヒメツリガネゴケの受精卵の第一分裂に必要なPpLFY転写因子は陸上植物の2倍体の多細胞体制獲得に関与しているかもしれないのでその相同遺伝子をシャジクモから単離した。さらにヒメミカヅキモの形質転換の条件を検討し、安定な株を単離できるようにし、ESTから得られた候補遺伝子の機能解析を進めている。これらの結果は、今後、さらに新型シーケンサーを用いたdeepシーケンシング、ゲノム配列の決定を行い、多細胞体制獲得の鍵が何であったかを解明する基盤となる。
