リパーゼ類のイオン液体中での酵素反応報告は多くなされているが、リン脂質代謝酵素に関しては報告例が全くなかった。そこでまず、PLD酵素がイオン液体中で安定して存在するか、8種のイオン液体を用いて検討を行った。各イオン液体と酵素溶液)in 20mM Tris-HCl)pH 7.4)をそれぞれ重量比で50%になるように混合し、30度にて3日間、攪拌および保温した。その結果、初日のPLD活性を100%とし比較を行った結果、何も加えない系では1日の加温にてPLD活性は約25%まで減少するのに比較して、BminPF6およびTMPATFSIと接触させた系では、3日間の接触によっても100%の酵素活性を維持していることが明らかとなった。接触させることで酵素活性が対照実験と比較して減少する系もあることから、両者のイオン液体はPLD酵素の安定性に寄与している可能性が高い。イオン液体中でのPLD酵素の安定性が確認できたので、次にこれまでのPLD酵素では酵素失活が激しいために顕著な反応生成物が確認できなかった、水相・n-ヘキサン系へのイオン液体の添加効果)酵素安定性向上効果)を検討した。その結果、アニオンがTFSI系のイオン液体では、対照実験と比較して約2倍の酵素安定性が確認できた。また、リン脂質はイオン液体には溶解しないと事前実験より確認していたが、OminBF4系では長期に静値することでリン脂質が溶解することも確認できた。
