われわれは二酸化チタンに超音波を照射することにより活性酸素種(ラジカル分子)が発生する現象を見出した。この現象をがん治療への新たな手法として応用することが本研究の目的である。本申請研究では(1)チノ二酸化チタン包括ターゲティングリポソームの開発、(2)創製した二酸化チタンと超音波照射法を培養細胞に適用することで、どのようながん治療の効果が得られるのか検討、そして(3)がん組織(がん細胞)におけるラジカル攻撃の分子レベルの挙動解析の3点について検討を行った。(1)項においては、センダイウィルスや特定のタンパク質をリポソーム表面に固定化した二酸化チタン包括リポソームの構築を完了した。更に、このリポソームを用いて、(2)項にて培養細胞との融合実験および、超音波照射による細胞死滅効果について検討を行い。二酸化チタンの効率的取り込みと、取り込んだ細胞における超音波照射による効果的な細胞死滅現象が確認できた。そして(3)項では、細胞の死滅機構にラジカルが関与していることが実証できた。この事より、二酸化チタン・超音波照射法によりラジカルが発生し、そのラジカルが細胞死を誘導していることが示唆できた。
