Note (General): 金沢大学がん進展制御研究所
本研究の目的はヒトがん組織において広範に発現し、悪性度・浸潤性との相関性が高い膜型マトリックスメタロプロテアーゼー1(MT1-MMP)活性制御分子・新規基質を検索し、その浸潤・転移に果たす役割を解析することにより、MT1-MMPを分子標的とした診断・治療法の開発へと発展させることにある。我々はこれまでにMT1-MMPが細胞運動、増殖に重要なExtreacellular-Signal Regulated Kinase(ERK)の持続的な活性化に必須の役割を果たすことを報告してきた。今回このシグナル伝達の中心分子であるFocal Adhesion Kinase(FAK)のリン酸化に果たすMT1-MMPの役割を検討した。細胞をファイブロネクチン、コラーゲン上にまくと接着斑が形成されFAKの397番目のTyrがリン酸化されるが、MT1-MMP活性を阻害すると、このTyrリン酸化が過剰に蓄積し接着斑が肥大化することを見出した。すなわちMT1-MMPによる細胞と細胞外基質との接着制御は接着斑形成のターンオーバーを促進し、結果として持続的なシグナル伝達を可能にすることが明らかとなった。また、がん組織で通常認められるMT1-MMPによるMMP-2のプロセシングについて、プロセシングされたMMP-2の酵素活性を検討した。MT1-MMPによるMMP-2のプロセシングにはその阻害因子であるTIMP-2が必須であるが、プロセシングを受けたMMP-2が実際に酵素活性を有するためには、きわめて限られた範囲内のTIMP-2濃度でなければならないことを見出した。この知見はTIMP-2のがん組織での果たす役割を考慮するうえで重要と考えられた。
