成熟動物の小脳プルキンエ細胞への興奮性シナプスでは、神経細胞の電気的な活動を修飾するだけで形態学的な変化を伴うシナプス結合の変化が起こる。昨年度までの研究から、AMPA型グルタミン酸受容体がシナプスに輸送されないStargazerマウスでは、登上線維が機能的に弱化しており、シナプス前終末からの伝達物質放出量が減少していることが明らかにされた。すなわち、AMPA受容体が登上線維の機能維持に必須であることが考えられる。今年度は、さらにプルキンエ細胞内において登上線維の機能維持に関わる因子を探索するため、プルキンエ細胞樹状突起棘(スパイン)内の細胞内ストアーからのカルシウム放出に異常が見られるDilute Neurological (DN)マウスの解析を行った。DNマウス小脳からスライスを切り出し、スライス表面上のプルキンエ細胞からホールセルパッチクランプ法で記録を行い、シナプス電流を記録した。その結果、登上線維応答振幅が正常に比べて有意に減少していることが分かった。また、シナプス前終末からの伝達物質放出過程の変化を反映するpaired-pulse depressionがDNマウスにおいて起こりやすくなっている傾向が見られた。また、形態学的な解析から、登上線維のシナプス形成領域が減少していることが分かった。これらの結果は、成熟動物においてAMPA受容体阻害剤(NBQX)を長期投与したマウス、及びStargazerマウスにおいて見られた登上線維の機能的弱化の症状に酷似しており、プルキンエ細胞内において神経活動により誘発される細胞内ストアーからのカルシウム放出が登上線維の機能的維持に関与している可能性が示唆された。
