難治性角結膜疾患の有効な治療法である角膜輪部移植術の成績をさらに向上させるには、角膜輪部の生理的、又は病的状態での機能の更なる解明が必要である。そこで、研究代表者らは角膜輪部の機能の中でも生理的に重要である痛みや、栄養因子の放出を行っている神経やシナプス小胞の局在、機能を調べることを目的として本実験を行った。まず、角膜幹細胞の同定のためにp63に対する抗体を使用してそのラットの眼表面における局在を研究中した。その際、眼瞼、結膜、角膜を一塊として摘出して、免疫染色を行った。その結果、ラットにおいてp63は角膜輪部での発現はそれほど多くはないが、結膜に散在的に存在していることが判明した。このことにより、p63が結膜の幹細胞のマーカーにもなりうる可能性が示された。また、眼瞼においては睫毛の毛根においてp63が多く発現されていることを見出した。このことから、p63の毛根の幹細胞のマーカーとしての役割について現在も検討している最中である。さらに、研究実施計画書に基づき、正常マウス、ラット、ウサギを用いて神経関連遺伝子(nerve growth factor, brain-derived neurotrophic factor, neurotrphin-4,cilialry neurotrophic factor, glial cell line-derived neurotrophic factor, neurophilin)とシナプス小胞関連タンパク質(Complexin2,Doc2,Munc-18,Rabaptin-5,Rabphilin-3A, rSec8,SNAP-25,Synapsin, Synaptotagmin, Syntaxin 4,Syntaxin 6)に対する抗体を用いて角膜輪部(角膜、結膜、眼瞼を含む)に対する免疫染色をおこなった。その結果、nerve growth factor, brain-derived neurotrophic factor, neurotrphin-4,cilialry neurotrophic factor, glial cell line-derived neurotrophic factor, neurophilinなどの角膜神経における発現が見出された。この実験結果について、現在再度確認中である。
