肝組織からの分離された細胞を包括的１細胞遺伝子解析することにより、肝臓を構成する細胞種の同定と機能解析、さらには疾患肝からの解析により病態を形成する細胞集団の特徴を明らかにした。その結果、肝細胞、胆管細胞の上皮細胞のみならず、免疫担当細胞や間葉系細胞の多様性がみられ、肝疾患の病態を形成する細胞種は疾患毎に多彩であり、また疾患を特徴付けるものであった。さらに、従来から肝病理組織でみられる組織所見について細胞集団データから解析し、肝病態を形成する各組織所見の意義について解明することができた。今回の先駆的な解析法により、正常肝および疾患肝における細胞社会の詳細を明らかにすることができた。
The novel analysis of single cell analysis using human liver tissues revealed the identification and function of each cells composing normal and diseased livers and characterized the cell populations organizing various pathological conditions. Immune cells and mesenchymal cells as well as liver resident epithelial cells (hepatocytes and cholangiocytes) consist of varied cell populations and its heterogeneity could characterize each liver diseases. Moreover, the pathological significance of histological findings found in various hepatobiliary diseases could be clarified from the aspect of the comprehensive gene expression in each constituent cells.
