高齢者では加齢に伴う筋量の減少に加えて、疾病などで活動性が低下すると筋萎縮が急速に進行するとされている。高齢者の骨格筋の反応が若年者と異なるのであれば、不活動期間中の運動療法も高齢者により適した方法を検討する必要がある。本研究では不活動状態下で施行した荷重練習の効果について、加齢による反応の違いを骨格筋線維の核動態および組織化学的分析から検索した。方法は後肢懸垂モデルによる廃用性筋萎縮後のラットで荷重を施行した。3ケ月齢、7ケ月齢、14ケ月のラットで、それぞれ、a)懸垂群(懸垂飼育)、b)荷重群(懸垂飼育し、1週間後より1日1時間後肢を下ろして荷重)、c)対照群(通常飼育)の3つの実験群を設け、3週間の実験終了後に下肢筋を採取し、凍結切片を作成した。核動態に関して、筋線維の衛星細胞の増殖活動は3ケ月齢では対照群に比べて懸垂群、荷重群では減少しており7ケ月齢、14ケ月齢では減少した。アポトーシス核の検索では、加齢に伴い増加していたが荷重群と懸垂群との違いは明らかではなかった。筋線維横断面積(MCSA)の比較では、すべての月齢で対照群に比べて懸垂群、荷重群のMCSAは減少していた。荷重練習は8ケ月齢では比較的有効であり、3ケ月齢では懸垂群との間に違いは生じなかった。一方、14ケ月齢のTypeI線維において、荷重群のMCSAは懸垂群よりも若干低値であり、自重による荷重であってもその反応は月齢により異なっていた。
In the skeletal muscle of the elderly there is a decrease in the muscle mass as well as a decline in muscle strength. The atrophying process in the absence of activity in the elderly may differ from that in the young due to age-associated changes in their skeletal muscles. It is necessary for physiotherapy to review an adequate method by the elderly during an inactivity period. In this study, nuclear kinetics and histochemical analyses of hindlimb muscle of each rat were conducted to determine the effect of weight healing (WB) on disuse atrophy would differ with advancing age.3-, 7-, 14-month-old male rats were assigned to two experimental group and one control group for a 3-week experiment. HS group had their hindlimb kept in suspension. HS+ group were also suspended but subjected to loading of the hindlimb muscle through WE hr one hour per day that began one week after the commencement of the experiment The control group was subjected to no suspension. We obtained hindlimb muscle after experiment termination and made cryosection.We compared increase in planting activity of a satellite cell with control group at 3-month-old and decreased in HS group, HS+ group. It decreased at 7-month-old and 14-month-old. The proliferative activity of apoptosis nuclei increased with age. However, there was no marked difference between HS and HS+ groups.Muscle fiber cross-sectional area (MCSA) of HS group and HS+ group decreased than control group regardless of age. WB was relatively effective at 7-month-old. At 3-month-old, there was no marked difference between HS and HS+ groups. In type I fibers at 14-month-old, MCSA in HS+ group was slightly lower than that in the HS group. The response to own-weight loading depended on age.
