Note (General): 金沢大学医薬保健研究域医学系
新たなシナプス伝達機構の解明を目的として、小胞の形質膜結合に関与すると推定されるExocyst複合体に注目し関連遺伝子の線虫ホモログ、Sec5,10,15及びExo70遺伝子のクローン化を行った。その結果を基に、発生段階におけるそれぞれの遺伝子転写産物の定量的PCRによる遺伝子発現レベルの検討を行った。Sec5,10及びExo70遺伝子の発現はEmbryo期で最も高く、ステージ移行とともに減少していた。このことは神経系の発生にもこれら遺伝子が関与すると推測される。Sec15遺伝子は、今回、新たに1種の転写産物の存在が明らかになり、これまでに6種が判明している。基本となるcDNAは24個のエキソンからなるが、その他の転写産物はオルターネイティブスプライシングよりなっていた。いずれの転写産物もエキソンが短いことからPCRの性質上定量的解析にはいたっていない。しかし、この遺伝子は神経系と腸に発現することから分泌系に機能していることは確実である。クローン化したSec3,Sec5,Sec10,Sec15,Exo70,Exo84遺伝子cDNAをHis-tag又はGST-tagを有するタンパク発現ベクターに組込み、大腸菌内で大量発現させた。発現させたタンパク質をGlutathion Sepharoseカラム或いはNi^<2+>カラムを用いて精製を試みたが、ほとんどのタンパク質は不溶性であったことから、さらに可溶性のタグを持つベクターにサブクローニングし、発現、精製を行った。精製を行ったSEC10とUNC-18との結合実験からSEC10はUNC-18と相互作用することが判明した。これによりSEC10は神経伝達物質の放出に関与すると考えられた。
