嗅上皮に存在している嗅神経細胞は多くの神経組織に分化しうる神経幹細胞を含んでいることが示唆されている。しかし現時点では嗅神経細胞、基底細胞の経代培養ならびに再生を目的とした神経組織への移殖は確立されていない。我々はmatrix metalloproteinase-2(MMP-2)が嗅上皮基底細胞から未熟な嗅細胞に発現していることを初めて報告した。MMPsは間質を分解する酵素機能以外に細胞の分化、増殖などに関与していることが明らかにされつつある酵素である。このMMP-2の嗅上皮での発現を遺伝子レベルで明らかにする為、In situ hybridizationにてmRNAの組織での局在を、RT-PCRにて嗅神経細胞の変性と再生時におけるMMP-2のmRNAの定量的変化を観察した。その結果、mRNAも蛋白と同様の局在と変化を示すことがわかった。また近年クローニングされた接着因子であるSgIGSFが嗅上皮にも発現していることを発見した。SgIGSFはImmunoglobulin-Superfamilyの一つとして2001年にみつかった接着因子でありNCAMとの類似性からneuriteの進展やシナプスの可塑性に関与している可能性がある。SgIGSFはMMP-2と同様、未熟な嗅神経細胞から基底細胞に発現していた。嗅神経細胞に変性と再生を起こすモデルではSgIGSFは再生の時期に増加し、これはMMP-2と反対の動きであった。培養嗅細胞でのMMP-2の発現を観察すべく当教室にて細胞培養系を立ち上げた。上記の実験に加えてLipopolysaccaride(LPS)刺激による嗅上皮の変化をin vivoで観察した。LPS点鼻連続14日間刺激によりラット嗅上皮はその厚さは変化しないものの、アポトーシスが誘導された。その変化は可逆的で点鼻終了後1日目が最も多くのアポトーシスにおちいった細胞を認め、7日目には対照のレベルにまで回復していた。Bax、bcl-2の免疫染色、Caspase-3の免疫染色でLPS点鼻後は陽性細胞が対照より増加しており、アポトーシスはこれらの系を介することが明らかとなった。
