本年度も種々の新しい知見が得られたが、その中でも進展の著しかった転移抑制物質スクリ-ニング法を中心に記述する。転移抑制物質のスクリ-ニング法として鶏卵法の有用性をさらに追求すべく、転移抑制試験に使用可能なヒト腫瘍細胞の探索を行なった。25種のヒト腫瘍培養株について鶏卵胎児における実験転移能を調べた結果、骨軟部悪性腫瘍4株(HTー1080,MNNG/HOS,SKーESー1,MGー63)、胃癌(KKLS)、肺癌(PCー8)、結腸癌(COLO320)、喉頭癌(HEpー2)、及び膀胱癌(Tー24)は鶏卵胎児の肝及び肺に転移することが明らかとなった。従って、これらの転移性ヒト腫瘍を用いることにより腫瘍型に応じた転移抑制物質の探索が可能となった。癌遺伝子は正常細胞の癌化のみならず、癌の転移・浸潤能の発現にも関与することが既に知られている。癌の転移形質発現に関与する癌遺伝子を標的とした転移抑制物質のスクリ-ニング系の確立を目的に、鶏卵法を用いて、癌遺伝子で形質転換したマウス胎児細胞株NIH/3T3の転移実験を行なった。具体的には癌遺伝子Haーras、Kiーras、src、abl、及びfosで形質転換したNIH/3T3細胞を受精鶏卵漿尿膜上の血管内より移植し、鶏卵胎児の肝及び肺における転移細胞を我々が開発したPCR法を用いる転移腫瘍検出法により定量的に検出した。その結果、これらの形質転換細胞はいずれも鶏卵胎児臓器に転移し、中でもras遺伝子(vーHaーrasおよびvーKiーras)形質転換NIH/3T3細胞は極めて強い転移性と転移臓器における高い増殖能を示した。以上の結果は、他の実験動物を用いた転移実験の結果と一致し、鶏卵法が転移実験系として有用であることが確認された。鶏卵法および癌遺伝子形質転換細胞を用いることにより転移関連癌遺伝子に照準を合わせた転移抑制物質の開発研究が期待されると共に、転移機構解明に有用な実験系となることが期待される。
