HBV持続感染と肝癌発生との関連を1)肝癌及び慢性肝炎試料のHBV DNAの組み込み様式の構造解析。2)HBVゲノムの転写及び複製制御のシス情報の解析とトランスに結合する蛋白の同定、3)ウイルスX蛋白のtrans-activatorとしての可能性、4)動物モデル系の肝発癌とウイルスの関与の検討を行なった。主要な成果として1)培養肝細胞のHBV感染系を用い、感染後早期に再編成されたHBV DNA組み込みを検出した(落谷班員)。慢性肝炎試料で癌化に先だち多様な再編成を受けたHBV DNAの組み込みの存在が示された(小池班員)。これらの結果はHBV DNAの組み込み、または再編成が肝発癌と関連するとの作業仮説を支持しない。多中心性肝癌の機構、癌の進展機構の解析に組み込みの様式を癌細胞のクローナルなマーカーとして解析する方向が進展し(小林、安井班員)、動物モデル系でのウイルス増殖と癌試料のウイルスDNAの組み込み構造の解析が行われた(小俣、吉川班員)。2)エンハンサーの活性化と結合蛋白の検討を進め、c-jun/fos関連蛋白が活性化に関与し、更にX蛋白の標的領域であることが示された(村上)。X蛋白がmyc遺伝子上流制御領域のtrans-activatorとして機能することが示唆された(小池班員)。検討を進めるべき課題として、1)X蛋白が異なったシス情報をtrans-activationし、ウイルス増殖のみならず宿主の増殖関連遺伝子の活性化により細胞増殖をもたらし、癌化の前提となる細胞集団のexpansionに関与する可能性が提示されている。X蛋白のtrans-activation機能の解析をHBV及び宿主のXREと結合する制御蛋白と解析とXのmidulation機構の検討が重要となった。2)肝癌発生にはHBV感染後長期の潜伏期が疫学的に示されている。今後ヒト肝癌とウイルスの関与を理解する上でhypernodule、初期癌、進行癌の過程の特性を癌関連遺伝子の活性化、suppressor遺伝子の欠損等の検索が必要となった。
