Note (General): 金沢大学医薬保健研究域医学系
活性化血小板から放出される生理活性リゾリン脂質、スフィンゴシン-1-リン酸(S1P)は、マウスB16メラノーマ細胞において内因性に発現するG蛋白共役型S1P受容体、S1P2/Edg5サブタイプを介し、in vitroにおいて細胞遊走と細胞外マトリックス浸潤を抑制する。本研究では、B16細胞のC57BL/6マウス皮下接種によるメラノーマ腫瘤形成が、S1Pの連日局所投与により用量依存性に著明に抑制されることを見出した。病理組織学的検討ではS1P投与群においてがん組織の線維性被膜による隔絶と周辺組織への浸潤の欠如が認められた。In vitroにおいてS1PはB16細胞の増殖を抑制しなかった。S1P2/Edg5ノックアウトマウスを用いた検討から、S1Pの抑制効果は宿主側間質細胞ではなくB16細胞の発現するS1P2/Edg5を介し発揮されるものと考えられた。B16細胞にS1P2/Edg5受容体を強発現させ皮下接種するとS1Pの抑制効果に対しより感受性となった。一方、B16細胞にS1P1/Edg1受容体(in vitroにおいてGiを介し細胞遊走・浸潤を促進する)を強発現させると、メラノーマ腫瘤形成がS1P依存性に増強された。これらの知見からin vivoにおいてS1Pはがん細胞が発現する受容体サブタイプ特異的にがんの浸潤、腫瘤形成を促進あるいは抑制することが明らかとなった。さらに、培養血管内皮細胞へのB16細胞の接着・伸展がS1Pにより抑制され、これはS1P_2/Edg5選択的阻害剤により解除されることを見出した。これはS1P_2/Edg5を介する血行性肺転移抑制の分子機構の一端を担うと考えられる。
