我々は1999年にhTERTプロモーターのクローニングに成功して以来、hTERT発現の分子機構を解析してきた。hTERT転写活性化因子としてc-Myc/MaxやSp1,ER, EWSなどいくつかの因子を見いだしたがこれらの因子では癌特異的なテロメレース活性化機構を説明するのは不十分であり、他に重要な機構が存在すると推定される。そのひとつが正常細胞におけるhTERT抑制機構である。今回の研究ではERM (Enhanced Retroviral Mutagenesis) systemを用いてhTERT転写抑制因子のクローニングを試みた。HeLa細胞を用いてtTA細胞を樹立し、hTERT promoter-GFPレポーター遺伝子を導入した。TREおよびsplicing donar sequenceを含むプロモーター領域をLTR下流につないだレトロウイルスベクターを上記tTA細胞に導入し、GFP発光強度が著しく減少した細胞集団をFACS解析にて回収した。さらに2次スクリーニングでテトラサイクリンを添加し、GFP陽性転化する2クローンを回収した。現在これらのクローンでレトロウイルス下流に存在する遺伝子の単離を進めている。さらにhTERT転写活性化に関わる新たな転写因子として低酸素環境下に誘導されるHIF1がhTERT core promoterのE-boxに直接結合し、低酸素化で腫瘍細胞のhTERT転写活性化に重要な役割を演じていることを明らかにした。
