われわれは二酸化チタンに超音波を照射することにより活性酸素(ラジカル分子)が発生する現象を見出した。この現象をがん治療への新たな手法として応用することが本研究の目的である。本研究で提案する超音波照射法にてラジカルを発生させれば、皮膚組織や臓器の表面だけでなく深部のがん組織においても治療が可能となり、非侵襲的で根治性の高いがん治療法を開発できる。そこで本申請研究では(1)二酸化チタン表面に生体認識分子を配向化し、血流中を移動できるナノ構造化の検討(2)創製した二酸化チタンと超音波照射法を担がん動物に適用することで、どのようながん治療の効果が得られるかの2点について検討を行った。(1)項においては、中心に磁性粒子を有する機能性二酸化チタン微粒子の構築が確認でき、構築微粒子の光触媒活性をメチレンブルー分解にて評価した結果、光触媒活性を有することも確認できた。更に、チタン・ナノ粒子内包リポソームも構築でき、がん細胞との融合実験に用いることが出来るレベルであることが明らかとなった。(2)項では、腫瘍細胞の生存率は超音波照射時間に依存して減少する事が明らかとなった。超音波照射のみでも腫瘍細胞死滅の効果はみられるものの、二酸化チタンを添加することでより効果的に腫瘍細胞を死滅できることが明らかとなった。
