DNAにナンセンス変異が生じている場合や、DNAの組換え、スプライシング異常などによって、結果的にmRNA上のアミノ酸をコードする配列に新たな終止コドンが生じることがある。この変異を持つmRNAを選択的かつ積極的に分解する機構をNMD(Nonsense-mediated mRNA Decay)という。スプライシングに伴ってExon Junction Complx(EJC)が形成され、NMDに必要なUpf複合体がmRNAに結合するための足場となる。EJCを介してmRNAに結合したUpf複合体は、その上流にあるナンセンスコドンを翻訳停止に伴って検出し、mRNAを分解に導く。よってNMDにおいて、Upf複合体がmRNAへ結合するためにはスプライシングが必要であり、また変異を認識するためには翻訳反応が必要である。これまでの研究で、NMDは、スプライシングがおこったCap Binding Complex(CBC)結合mRNA上での翻訳過程を通して変異を持つmRNAを選別して分解することが明らかとなった。本研究では、細胞から精製したCBC結合mRNA上にEJCとUpf複合体がmRNAを介して結合することを明らかにし、NMDの標的となるmRNA-蛋白質複合体のモデルを提示した。さらにCBC結合mRNAを基質として、NMDを試験管内で再構築することを目的として以下の実験を行った。FLAG付加されたヒトCBC発現プラスミドを細胞に導入し、安定に発現させた。この細胞についてFLAGに対する抗体を用いた免疫沈降を行い、FLAG付加CBCがmRNAキャップ構造に結合したmRNA-蛋白質複合体を回収することを試みた。
