転写やmRNAプロセシングといった核内事象を連携(カップル)させている分子機構を明らかにするアプローチとして、精製した二種類のリン酸化及び非リン酸化フォームのRNAポリメラーゼII(Pol II)のin vitroスプライシング反応に対する影響を解析した。その結果Pol II最大サブユニットのカルボキシル末端領域(CTD)が高度にリン酸化したPol IIはスプライシング反応を強く促進し、一方非リン酸化フォームPol IIは反応を逆に抑制することを見い出し(Hirose et al. Genes & Dev.1999)、Pol IIがmRNAプロセシング装置と物理的に相互作用出来るだけでなく、CTDのリン酸化調節を介してプロセシング反応を機能的に制御できる可能性を生化学的に示すことが出来た。更にPoll IIが、CTDのリン酸化調節を介して、転写反応のみならずmRNAプロセシング等の他の核内事象に如何なる分子間相互作用を通じて関わっているかを明らかにするために、リン酸化CTDに結合する新規蛋白質の同定をFar-western法を用いた発現クローニングによって試みた。これまでの解析から、リン酸化CTDに結合する候補蛋白質としてヒトの新規蛋白質PCIF1(Phosphorylated CTD Interacting Factor 1)及び既知のヒト核蛋白質Pin1を同定した。これらの蛋白質のリン酸化CTDとの結合責任領域をFar-western法及びGST-pull down法で同定した。新規蛋白質PCIF1については、flag-PCIF1叉はGFP-PCIF1をトランスフェクトした細胞を用いた免疫共沈実験、及び細胞内局在解析実験より、内在性のリン酸化RNAポリメラーゼIIとPCIF1の細胞内における特異的会合を確認した(未発表データ)。
