RNAポリメラーゼII(RNAP II)により転写される遺伝子の転写調節は、基本転写因子(TFIID,TFIIB,TFIIE,TFIIFとTFIIH)、転写調節因子、それらの間を仲介する仲介因子(メディエーター)の相互作用により制御されている。転写開始から伸長への移行過程で、RNAPIIのCTD(carboxy-terminal domain)はTFIIHのCTDキナーゼ(Kin28タンパク)によりリン酸化される。最近酵母において、いくつかの遺伝子の転写には、TFIIE、Kin28およびCTDが必要ないことが示されている。本研究では、このようなTFIIEとCTD/Kin28に対して非依存的な遺伝子が、どのように転写されるかについて検討した。CTD/Kin28は、heat shock遺伝子およびCUP1(銅メタロチオネイン)遺伝子の転写活性化には必要ない。TFIIEはHsf1(heat shock factor)によるheat shock遺伝子の転写活性化には必須であったが、銅によるCUP1の活性化には不要であった。CUP1の転写には、Hsf1とAce1(銅による活性化因子)の2つが関与しているが、TFIIE非依存的になるのは、両方の活性化因子が存在する時に限られた。つまり、CTD/Kin28非依存的なHsf1による転写活性化において、TFIIEはCTDのリン酸化以外の過程に必須であり、このTFIIE要求性もAce1により回避されると考えられる(BBRC,261:734-739)。heat shock遺伝子のKin28非依存的な転写活性化の機構について検討した結果、Hsf1のC-末端付近にある転写活性化ドメインが、非依存的な転写を担っていることが明らかになった。また、heat shock遺伝子とCUP1遺伝子の転写活性化に必要な仲介因子を検索した結果、メディエーターの構成タンパクであるGa111,Med2,Pgd1およびSin4が、仲介因子として機能していることが示唆された(論文準備中)。今後、このメディエーターを介したTFIIE/Kin28非依存的な転写開始複合体形成の機構と、転写の開始から伸長反応への移行制御について、さらに検討が必要である。
