B型肝炎ウイルス(HBV)X蛋白(HBx)は肝細胞がん発症への関与が示唆されている。我々は、RNAポリメラーゼサブユニット5(RPB5)がHBxの宿主標的であることを見い出し、HBxがRPB5、転写基本因子TFIIBとの3者の相互作用し、転写補助活性化能(coactivator)を示すことを見い出した(J.Biol.Chem.,1998)。更にHBxに拮抗する宿主蛋白として、転写抑制補助能(corepressor)を示すRMP(RPB5-mediatingprotein)を同定した(Mol.Cell,Biol.,98)。本年度の研究成果として、1)HBxが標的とするRPB5とTFIIBの相互作用の生物的な機能の解析を酵母細胞系で検討した。その結果、TFIIBのN端及びC端領域は、酵母とヒト間で互換性を示すことを見い出した。またyRPB5V110G(ヒトのcounterpartはV105に相当する)の高温度感受性変異は、yTFIIB又はヒトと酵母のキメラTFIIB(hTFIIB(aa1-47)/yTFIIB(aa58-339)の過剰発現によって形質抑制(phenotypic suppression)を受ける結果を得た。この結果は、酵母細胞とヒトの系でRPB5とTFIIBの機能的な相互作用が認められることが示された。2)RMPの機能的な拮抗因子と推定されたRMPは、細胞質と核に共に見い出された。転写を負に制御する機能にはRPB5結合部位以外に、RMPのC端領域が必要であり、その領域に存在する核移行シグナル(NLS)が必須であった。更にRMPのN端側にRMPの核への移行を干渉する領域が存在することを見い出した。細胞質への滞留に関与する配列はaa81-118内に限定され、この領域はcoiled-coil領域とほぼ一致した。細胞質に滞留する分子機構と核内への移行を促進する因子については、現在検討を進めている。
