レプチンはobgeneからつくられる蛋白質であり、ここ数年の間に生理薬理学的作用の一部が解明されてきた。レプチン欠損マウスの体温および熱産生量は正常マウスと比し有意に低くレプチンの抹消もしくは中枢投与で体温が上昇し熱産生量は増加する。これらの報告は、レプチンが体温調節に関与している可能性を示唆している。平成10年度にはレプチンが体温調節に強く関係することを報告した。本年度はレプチンの下流で働く因子を検討した。非拘束・正常ラットの皮下にシクロオキシゲナーゼ阻害薬のインドメタシンもしくは免疫抑制作用のあるデキサメサゾンを前投与し、一定時間を経た後ヒト-レプチンをラットの脳室内にマイクロインジェクションした。前投与後からの体温変化をテレメトリーシステムを用いて測定した。インドメタシンもしくはデキサメサゾンを前投与するとレプチンによる体温上昇は抑制された。また、インドメタシンはレプチンの摂食抑制に影響を与えないが、デキサメサゾンはレプチンの摂食減少を抑制した。上記の結果から、レプチンの体温上昇作用はプロスタグランジンやサイトカインを介していること、さらに、レプチンの摂食抑制はサイトカインを介していることが示唆された。
