多様なストレスにたいする細胞の応答機構における分子シャペロンの役割について明らかにするため、これらストレスによって活性化されてストレス応答に重要な役割を果たしていると考えられているNF-κB転写因子の活性化のシグナル伝達機構における分子シャペロンの役割について研究し、現在までに次のような成果を上げた。我々は前に免疫抑制剤FK506によりIκBαの分解を介してNF-κBの活性化が起こることを明らかにした(J.Clin.Invest.,1996)。NF-κBの活性化には、そのインヒビターであるIκBαのN末部のセリン残基の燐酸化とユビキチン化に依存したプロテアソームによる蛋白分解が必須であると考えられている。我々はFK506とFKBP分子シャペロンによるNF-κBの活性化ではIL-1/TNF-αの場合と異なり、N末部のユビキチン化部位の非依存性にプロテアソームによる分解が起こることを明らかにした。また、42番目のチロシン残基のアラニンへの置換には影響されないが、N末部のセリン残基の変異によって分解はブロックされた。しかし、燐酸化したN末部のセリン残基に対する抗体を用いたウェスタンブロティングや、in vitro kinase assayによるIκBαキナーゼの活性化の測定、等IκBαの燐酸化を検出できないという興味ある結果が得られた。今後FKBPがIκBαの燐酸化とプロテアソームによる分解にどのように関わっているのか明らかにしていく。
