1.アポ蛋白のmRNAの定量法の検討サーファクタントアポ蛋白AのmRNAの量は,相補的DNAオリゴマーを用いてNorthern blotting法をすることにより定量できることを確認した. アポ蛋白BとCはこの方法で定量できず,別の方法の検討が必要と考えられた.2.肺洗浄ラットに対するアポ蛋白A添加サーファクタントによる治療効果の検討肺洗浄により急性呼吸窮迫症候群を呈したラットに対して従来型サーファクタントとアポ蛋白Aを添加したものの両者を投与して治療効果を比較した. その結果,対照動物では動脈血酸素分圧(PaO_2)が100mmHg前後であるのに対して,従来型ではPaO_2が300mmHg台まで回復して,効果の持続は約3時間であった. アポ蛋白A添加によりPaO_2が400mmHg台まで改善し,効果は約5時間持続した. このことから,アポ蛋白Aの添加は治療効果を増強すると考えられた. 実験終了後に肺を摘出して凍結保存してあり,平成11年度にアポ蛋白AのmRNAの定量を行う予定である. なおSP-A添加サーファクタントの活性を安定させるために時間を要したが,重要な知見と考えられた.3.防水剤によるサーファクタントの活性障害に関する追加実験防水剤によるサーファクタントの活性障害は,防水剤の中の有機溶媒が原因でなく,フッ素樹脂が原因であることを気泡型表面張力計で発見した. この知見は防水剤の吸入による呼吸不全の原因解明のために重要であり,Intensive Care Medicineに印刷中である.
