がんが転移する際のリンパ管およびリンパ管新生の役割は,現在がん生物学の分野で真剣に研究されている.しかしながら,がん細胞の腫瘍からの流出と,リンパ節への集束を伴うがん細胞のリンパ管への移動のリアルタイム・イメージング技術の開発は,これまで充分ではなかった.そこで我々は,これまで開発してきた蛍光蛋白発現腫瘍細胞を用いることで,リンパ管中でのがん細胞の移動について観察をおこなった.具体的には,蛍光蛋白標識した細胞をヌードマウスの鼠径部のリンパ節に移植し,鼠径部のリンパ節からリンパ管を通って移動し,腋窩のリンパ節に到達するまでの過程を,皮弁を通して細胞レベルでイメージングすることに成功した.また特にこの観察には,オリンパスOV100小動物イメージングシステムが,リンパ管を移動するがん細胞のリアルタイム・イメージングに有用であった.このイメージングテクノロジーを用いて,リンパ管へ流出するがん細胞への圧力の影響について検討をおこなった.この検討から,細胞断片,細胞,および塞栓の数は圧力が増すにつれて顕著に増加し,流出した塞栓の最大直径もまた圧力が増すにつれて大きくなることが明らかとなった.これは,腫瘍の圧力は周囲のリンパ管に進入する腫瘍細胞数のみならず,塞栓の大きさにとっても重要であり,より大きい塞栓の方がリンパ管内で生き残り,転移を形成する可能性が高いものと考えられた.がん細胞のリンパ管内の移動をイメージングする技術は,これまであまり理解されていなかったリンパ節転移の重要なステップの可視化を可能とした.
