我々は、先にサイトメガロウイルスプロモーターの下流にRenilla luciferase(R-luci), HCVの5'NTR, Firefly luciferasc(F-luci)の順で遺伝子を組み込んだベクターを作成し、この遺伝子を恒常的に発現しているHuh-7由来細胞を樹立した。この樹立細胞では、R-luciは通常の真核生物の蛋白翻訳機構であるキャップ依存性に、F-luciはHCVの5'NTRによりIRES依存性に翻訳される。この樹立細胞を用いた検討でHCVの蛋白翻訳効率は細胞分裂期で高いことを見いだしている(Gastroenterology118, 152-162, 2000)。今回、インターフェロン及び二本鎖RNA(Poly I : Poly C)処理によりIRES依存性蛋白翻訳が特異的に抑制されることを見いだした。この作用メカニズムを解明するため、インターフェロンによる、2-5AS(oligoadenylate synthetase)活性の誘導、PKR(2本鎖RNA依存性プロテインキナーゼ)の活性化、及びHCV-IRESを活性化させる宿主蛋白であるLa protein、及びPyrimidine tract binding protein(PTB)の発現を検討した。インターフェロンの投与により2-5ASは著名に上昇したが、Poly I : Poly C投与では活性の上昇は顕著でなかった。PKRの発現はインターフェロン、Poly I : Poly C投与で上昇したが、トランスフェクションによるPKR発現過剰状態ではIRES活性の特異的抑制は見られなかった。一方、IRES活性化因子のLa proteinはインターフェロン、Poly I : Poly C投与で発現が著名に抑制され、PTB及びコントロールとして用いたアルブミンの発現には変化が認められなかった。また、La proteinの過剰発現により、インターフェロン、Poly I : PolyCによる特異的IRES活性抑制は消失した。以上より、La proteinの発現がインターフェロンの抗ウイルス活性に影響を与えていることが明らかとなった(Hepatology35, 199-208, 2002)
