癌抑制遺伝子PTEN(phosphaatase aand tensin homologue deleted on chromosome10)は悪性腫瘍の悪制度と相関して変異・欠質が認められる。PTENの活性は細胞膜への移行およびPTENタンパクの安定性により制御されていると考えられる。このPTEN活性の制御にはそのカルボキシル末端が重要な役割を担っており、PTEN活性の制御機構を解明するためにPTENのカルボキシル末端をバイトにしてYeast Two-Hybrid法にてスクリーニングを行った。(結果)既知の4回膜貫通型分子とアクチン結合分子が同定された。培養細胞を用いた過剰発現系にて4回膜貫通型分子、アクチン結合分子とPTENの結合が免疫沈降法により確認された。この4回膜貫通型分子はC-末端の細胞内ドメインを介してPTENと結合することがわかった。また、アクチン結合分子はアクチン結合部位とは異なる部位でPTENと結合することがわかった。PTENのPI3-K/Akt経路抑制作用に対する4回膜貫通型分子ととアクチン結合分子の効果を抗リン酸化Akt抗体によるウエスタンプロッティングにて検討した。その結果、PTENによるAktのリン酸化抑制は4回膜貫通型分子と共発現することで増強された。(考察)インテグリン/FAK経路は細胞増殖、細胞分化、細胞運動を制御していおり、癌細胞浸潤機構解明の鍵であると考えられる。PTENはこのFAKの397番目チロシンを直接脱リン酸化することでFAKの活性を抑制し、PIP3を脱リン酸化することでも細胞運動を抑制する。最近、PTENのカルボキシル末端は膜への移行と安定性を制御していることが明らかになり、PTENのカルボキシル末端結合タンパクがPTENの活性を制御していると考えられる。今回PTEN結合分子として同定された4回膜貫通型分子はインテグリンと結合し細胞運動を制御していること、アクチン結合分子は細胞骨格形成に関与していることが報告されている。これらの経路にPTENがどの様に関与しているのか、FAKのリン酸化、MMPの活性発現に関与しているのかを今後検討する予定である。
