神経軸索切断後も生存する神経細胞の形態学的特徴を研究するために、顔面神経の耳介・眼瞼・上顎・下顎の各分岐枝に逆行性蛍光ニューロトレーサーであるジアミジノイエロー(DY)を投与し、各分岐枝を経て各顔面筋特異的に投射している神経細胞を逆行的に標識した後、顔面神経軸索を頭蓋内の小脳橋角部で切断し、神経細胞の生存率を調べた。その結果、軸索切断後4週間の時点で下顎分岐枝投射細胞の生存率が最も低く(36.2±0.8%)、次いで上顎(58.2±1.3%)、眼瞼(66.5±1.1%)、耳介(70.3±0.8%)の順に高くなっていた。生存神経細胞の形態学的特徴を調べたところ、軸索長と軸索切断後の生存率は密接に関連していることが示唆された。この結果は、軸索切断後の神経細胞の生存のためには標的器官のみならず神経軸索構成細胞によって産出され、放出される神経栄養物質の量も重要であることを示唆している。また、免疫組織化学染色によって軸索切断後の神経細胞核に特異的に発現する物質の一種であるc-junの発現率を調べたところ、長い軸索を持つ細胞ほど発現率が高かった。このことから、神経栄養物質の欠乏条件下にあっては、c-junは神経細胞死に関与することが示唆される。
