研究代表者がこれまでに収集してきたヒト乳がん臨床検体由来patient-derived xenograft (PDX)を活用し、ゲノム遺伝子変異の蓄積によるがん組織の不均一性と、幹細胞性を維持するシグナルを指標とした機能的ヒエラルキーを統合させる研究を行った。シングルセルRNAシークエンスより、増殖能が低い親玉がん幹細胞様細胞がクラスターを形成する遺伝子の中から、親玉がん幹細胞と通常のがん幹細胞を区別できる膜タンパク質FXYD3が見いだされた。エクソームシークエンスより、がん幹細胞様細胞に濃縮されやすい遺伝子変異がこれまで見逃されてきたことを強く示唆している。
To integrate the heterogeneity in cancer tissues due to accumulation of multiple mutations in genome and functional hierarchy due to different differentiation states in cancer cells, we analyzed breast cancer patient-derived xenograft models and spheroid cells. Single cell RNA sequencing following clustering analysis revealed that there are at least 2 clusters: proliferating cell cluster and non-proliferating cell cluster. We found a membrane protein FXYD3 among the highly expressing genes in the latter cell cluster and demonstrate that expression of FXYD3 is useful for distinguish the 2 populations. Furthermore, we discover novel mutations in genes that are not identified by bulk tissue analysis in cancer stem cell populations.
