一細胞から多様な上咽頭癌組織構築過程について解析した。１）ヒト上咽頭癌細胞ではなく癌組織を有する上咽頭癌異種移植モデルマウスレパートリーを作製し、これまで困難であった腫瘍内癌細胞多様性の解析にを行った。内因性免疫APOBECはEpstein-Barr ウイルスのみならず、宿主DNA並びにミトコンドリアDNAに変異を導入し、上咽頭上皮細胞が癌幹細胞化した後、さらに多様に分化した癌細胞集団を形成する。2）NF-κBの制御により正常細胞の細胞競合能を増強し、オートファジー阻害を応用した新規治療法の有望性を確認し、次世代の上咽頭癌研究ならびにオートファジー制御による治療法開発の礎を構築した。
We developed patients derived xenograft of nasopharyngeal carcinoma (NPC) mice model and applied to analysis of heterogeneity in NPC tumor. APOBEC family that induce DNA mutations not only in EBV-DNA but in both host and mitochondria DNA, eventually, produce cancer stem cell and progression of heterogeneity of malignant cells. These results indicate that mutation inducing property of intrinsic immunity factor act double edge of sword for initiation and progression of NPC. Both promotion of cell competition potential in tumor-neighboring normal epithelial cell by modulating NF-kB, and inhibition of autophagy by chloroquine, are novel and promising treatment strategy for NPC.
