高速AFM/光ピンセット複合装置に特化した制御・解析プログラムを新規に開発し、これら二つの手法を同期して制御可能なシステムの構築を行った。更にモデル試料系として、長鎖ヘアピンコンカテマーアッセイの合成手法の開発にも成功した。こうして構築した実験系を用いて、ヘアピンDNAのアンフォールディングおよびリフォールディングするダイナミクスの可視化に成功した。更に、ヘアピンの根元にある不連続サイトを核としてssDNAがアンピーリングする様子や二次構造の形状が時々刻々と変化する様子の可視化にも成功した。これにより、生体フォールディング研究における大きな一歩を踏み出すことができた。
I successfully developed high-speed atomic force microscopy combined with optical tweezers via the development of control and analysis software that is capable of synchronous control of these two techniques. Furthermore, I also succeeded in developing a simple method for synthesizing a long-chain hairpin concatemer assay. By means of these newly developed systems, I successfully visualized submolecular-scale unfolding and refolding dynamics of the hairpin structures induced by an external force. The obtained results also indicated that a transitional change of bistable secondary structures over time as well as an unpeeling of ssDNA that is nucleated from a discontinuity at the hairpin construct. These achievements were definitely a giant step in the biological folding research.
